Why it matters: GLP-1 drugs are being studied far beyond weight loss, and any real effect on aging biology would be significant but this is an early, indirect signal in a specific group, not evidence that anyone lived longer.
Semaglutide, the drug in Ozempic and Wegovy slowed several markers of biological aging in the first randomized human evidence to test the idea. Two caveats belong right at the top: this was a post-hoc, exploratory analysis of a previously completed trial, and it was done in adults with HIV-associated lipohypertrophy so it can't be generalized to healthy people. The underlying study was a 32-week, double-blind, placebo-controlled phase 2b trial; epigenetic aging was analyzed in 45 semaglutide and 39 placebo participants. "Epigenetic clocks" estimate biological age from chemical marks on DNA. After adjustment, semaglutide significantly lowered several clocks versus placebo PhenoAge by 4.9 years, PCGrimAge by 3.1 years, and the DunedinPACE pace-of-aging by about 9%. Most important limitation: slowing a molecular clock is not proof of longer or healthier life, the researchers themselves call it an early signal, shown only in people living with HIV, not a verdict.
